1.一種固相多肽合成Exenatide的制備方法,其特征在于,包括如下步驟:
(1)制備Fmoc-Ser(tBu)-樹脂
稱取Rink?Amide?MBHA樹脂100克,裝入SHYM-25多肽合成儀中,用1000mlDMF浸泡30分鐘,加入Fmoc-Ser(tBu)-OH?58g,HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干;
(2)制備Fmoc-Pro-Ser(tBu)-樹脂
在步驟(1)的Fmoc-Ser(tBu)-樹脂中,加入脫帽試劑800ml,25℃反應30分鐘,抽干,用DMF、甲醇、DMF各洗滌數次,抽干,加入Fmoc-Pro-OH?52g、HBTU?58g、HOBT?21g,用1000ml接肽試劑溶解混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次抽干,獲得Fmoc-Pro-Ser(tBu)-樹脂;
(3)制備Fmoc-Pro-Pro-Ser(tBu)-樹脂
在步驟(2)的Fmoc-Pro-Ser(tBu)-樹脂中,加入脫帽試劑?800ml,25℃反應30分鐘,抽干,用DMF洗滌,抽干,加入Fmoc-Pro-OH?52g、HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干,獲得Fmoc-Pro-Pro-Ser(tBu)-樹脂;
Fmoc-Pro-OH的摩爾數為樹脂的2~5倍;
(4)制備Fmoc-Pro-Pro-Pro-Ser(tBu)-樹脂
在步驟(3)的Fmoc-Pro-Pro-Ser(tBu)-樹脂中,加入800ml脫帽試劑,25℃反應30分鐘,抽干,用DMF洗滌,抽干,加入Fmoc-Pro-OH?52g、HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解的混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干,獲得Fmoc-Pro-Pro-Pro-Ser(tBu)-樹脂;
Fmoc-Pro-OH的摩爾數為樹脂的2~5倍;
(5)制備Fmoc-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂
在步驟(4)的Fmoc-Pro-Pro-Pro-Ser(tBu)-樹脂中,加入800ml脫帽試劑,反應,抽干,用DMF洗滌,抽干,加入Fmoc-Ala-OH?48g、HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干,獲得Fmoc-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂;
Fmoc-Ala-OH的摩爾數為樹脂的2~5倍;
(6)制備Fmoc-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂
在步驟(5)的Fmoc-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂中,加入800ml脫帽試劑,反應,抽干,用DMF洗滌,抽干,加入Fmoc-Gly-OH?46g、HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干,獲得Fmoc-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂;
(7)制備Fmoc-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂
在步驟(6)的Fmoc-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂中,加入800ml脫帽試劑,反應,抽干,用DMF洗滌,抽干,加入Fmoc-Ser(tBu)-OH?58g、HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干,抽干,獲得Fmoc-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂;
(8)制備Fmoc-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂
在步驟(7)的Fmoc-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂中,加入800ml脫帽試劑,反應,抽干,用DMF洗滌,抽干,加入Fmoc-Ser(tBu)-OH?58g、HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干,抽干,獲得Fmoc-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro-?Pro-Ser(tBu)-樹脂;

在步驟(11)的Fmoc-Gly-Gly-Pro-Ser(tBu)?-Ser(tBu)-Gly-Ala-Pro-Pro-Pro-Ser(tBu)-樹脂中,加入800ml脫帽試劑,反應,抽干,用DMF洗滌,抽干,加入Fmoc-Asn(Trt)-OH?90g、HBTU?58g,HOBT?21g,用1000ml接肽試劑溶解混合物,25℃反應1小時,抽干,分別用DMF、甲醇、DMF洗滌數次,抽干,獲得Fmoc-Asn(Trt)-Gly-Gly?-Pro?-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-?Pro-Pro-Ser(tBu)-樹脂;
(13)制備Fmoc-Lys(Boc)-Asn(Trt)-Gly-Gly-Pro-Ser(tBu)-Ser(tBu)-Gly-Ala-Pro-Pro?–Pro?-Ser(tBu)-樹脂

將Exenatide粗品溶于重量濃度為5%乙酸中,過濾,濾液經C18柱純化,流動相:0.2M醋酸鹽緩沖液∶乙腈為7.5∶2.5,體積比;流速為350ml/min;檢測波長為:280nm;用液相色譜儀跟蹤收集所需要的流出液,樣品峰合并后去鹽,凍干,獲得產品12.2g,總收率為9.7%;
所說的接肽試劑為:NMM:DMF=1:5~15,體積比;
所說的脫帽試劑為:PIP:DMF=1:2~5,體積比。
